In the Neuro Probe A3BP48 chemotaxis chamber, chemoattractants added to the lower wells act on cells in a suspension added to the upper wells. The lower wells are formed by clamping the middle plate to the bottom plate. A filter separates the upper and lower wells. In the Neuro Probe A3BP48 chemotaxis chamber, chemoattractants added to the lower wells act on cells in a suspension added to the upper wells. After incubation the migrated adherent cells on this filter are stained and counted, as in the widely used?AP48. In addition, a membrane can be placed on the bottom of the bottom wells to collect migrated nonadherent cells to be stained and counted. If there is no membrane in the lower wells, nonadherent migrated cells can be viewed through the clear 5.7mm thick bottom plate of the A3BP48 with an inverted microscope. The Neuro Probe three-tiered chamber has three precision-machined acrylic plates, assembly hardware, and two silicone gaskets. The clear flat bottom plate has hardware holes only. The middle plate has 48 through holes and six permanently installed hardware posts. The top plate has hardware holes and 48 through holes. Raised rims around the holes of the middle plate form seals on both sides when it and the two gaskets are clamped between the top and bottom plates. Before the migrated cells adhering to the middle filter can be stained and counted, the nonmigrated cells must be wiped off the top side of the filter. An accessory pack?(Stock # P48AP)?includes instruments for this purpose. The A3BP48 is designed for use with polycarbonate filters, although cellulose nitrate filters can be also be used. Accompanying each chamber is an?A3BP48 protocol?for its use and care.
