In the Neuro Probe AC48 chemotaxis chamber, chemoattractants added to the lower wells act on cells in a cell suspension added to the upper wells. A pair of filters separates the upper and lower wells. After incubation the migrated cells on or in the filters are stained. With cellulose nitrate filters, one measures the depth the cells have reached within the filter material; with polycarbonate filters, one counts the cells that have migrated through the filter and adhered to its underside. The AC48 has special filter-retaining hardware designed to hold the two filters in place during assembly and disassembly of the chamber. The AC48 consists of precision-machined acrylic top and bottom plates, assembly hardware, and a silicone gasket. The bottom plate has 48 wells, each with 25μL volume and a diameter of 3.2mm. Corresponding holes in the top plate form the upper wells when the chamber is assembled. The chamber features eight button-head screws permanently mounted in the top plate, which hold the gasket and filters in place during assembly and disassembly. This filter-retaining hardware is positioned to accommodate two 35mm x 41mm pieces of filter material, each covering 24 wells. Narrow raised rims around mating surfaces of the top and bottom wells enhance the seal. The AC48 is designed especially for use with cellulose nitrate filters. Polycarbonate filters may also be used, but if that is your?primary?interest, we suggest you also consider the more commonly used AP48 chamber. Included with each chamber is an?AC48 protocol?detailing proper use and care of the chamber, as well as various protocols for?staining cellulose nitrate filters.
