The electroporation buffer system used with the Multiporator contributes towards increased efficiency. The use of these buffers (hypo and isoosmolar) with low conductivity prevents cell damage by reducing current flow. In hypoosmolar conditions, cells swell and the cell membrane becomes detached from the cytoskeleton, thereby enabling easier electroporation. Ideally, electroporation is carried out in hypoosmolar electroporation buffer; for very sensitive cells the buffer system can be diluted with isoosmolar electroporation buffer.
