Amyloid β/A4 Protein Precursor??? (586-595) (human, mouse, rat) also known as APP-derived inhibitory peptide (APP-IP), efficiently inhibited the activity of gelatinase A (matrix metalloproteinase-2 (MMP-2)) (IC?? = 30 nM), whereas its inhibitory activity towards membrane type 1-matrix metalloproteinase was much weaker (IC?? = 2 μM). The decapeptide ISYGNDALMP had poor inhibitory activity towards gelatinase B, matrilysin or stromelysin (IC?? > 10 μM). APP-IP is likely an active site-directed inhibitor of gelatinase A. Clarification of its mode of action might provide a lead for the design of novel anti-cancer drugs that block specific processes of tumor invasion and angiogenesis.
