Description	Product Specifications (Ala31) (Leu60) IGF-I: [Ala31][Leu60]IGF-I is an analog of human IGF-I with a substitution of Ala for Tyr at position 31 and a Leu for Tyr at position 60 of the human IGF-I sequence. Compared to IGF-I, [Ala31][Leu60]IGF-I has greatly reduced binding to the Type 1 IGF receptor and normal binding to acid-stripped human serum binding proteins (Bayne et al., 1990, J. Biol. Chem., 265, 15648-52). [Ala31][Leu60]IGF-I has very low affinity for the Type 1 IGF receptor and this affinity is lower than [Leu24]IGF-I or [Leu60]IGF-I. [Ala31][Leu60]IGF-I is therefore an extremely useful analog of IGF-I for investigating IGF actions largely independent of binding to the Type 1 IGF receptor. Source: [Ala31][Leu60]IGF-I is produced recombinantly as a fusion protein in E.coli using a patented expression system. The fusion protein containing [Ala31][Leu60]IGF-I is refolded, and after the leader peptide is removed, the [Ala31][Leu60]IGF-I product is purified to Receptor Grade using several chromatography steps. Purity: > 95% (as determined by HPLC and N-terminal sequence analysis) Molecular weight: Theoretical : 7507 daltons. Confirmed by Mass Spectometry. Appearance: Lyophilized white powder N-terminal sequence analysis: 5 residues > 95% single sequence Endotoxin (LAL): 500 ng/ml IGF binding protein assay - ED50 < 10 ng/ml State: Dried from 0.1 M acetic acid and stored under dry nitrogen at a slight vacuum (-25kPa) Reconstitution: Dissolve in dilute acid (e.g. 10mM HCl or 0.1M acetic acid), aliquot and store at -20°C. For lower dilutions (e.g. < 1mg/ml) inclusion of a carrier protein is recommended. Avoid freeze-thaw cycles. Storage/Stability: At least 2 years at 2-4°C (lyophilized).
